INTENDED USE
The Bovine Pregnancy (PAG) Rapid Test Cassette (Plasma/Serum) is a lateral flow immunochromatographic assay for the qualitative detection of Pregnancy-Associated Glycoproteins (PAG) in cattle’s plasma or serum.
SUMMARY
Pregnancy-Associated Glycoproteins (PAG) are macromolecules produced by placental tissue and released into the maternal circulation where they allow pregnancy diagnosis. They play an important role in the process of maternal reproduction and maintaining pregnancy.
Therefore, they can be monitoring cow reproductive activity by detecting of PAG. PAG has different characteristic changes as the cow's stage of reproductive physiology, through to detect the PAG levels can make an accurate diagnosis of cow in early pregnancy.
PRINCIPLE
The Bovine Pregnancy (PAG) Rapid Test Cassette (Plasma/Serum) is a lateral flow immunochromatographic assay. The test cassette has a testing window. During testing, sample is applied into the sample well on the cassette. If PAG present in plasma or serum specimen, react with anti-PAG antibodies coated colloidal gold particles in the test strip.
The mixture then migrates upward on the membrane chromatographically by capillary action and reacts in the test line region. If the specimen contains PAG, a colored line will appear in the test line region indicating a positive result. If the specimen does not contain PAG, a colored line will not appear in the test line region indicating a negative result. To serve as a procedural control, a colored line will always appear in the control line region indicating that proper volume of specimen has been added and membrane wicking has occurred.
THE BEST USE DATE
PAG immune response was significantly enhanced in cows at 24-28 days of gestation. The PAG reached (12.3±4.08) ng/mL at day 35 of gestation, then began to decrease, and continued to increase until day 56. Testing is recommended at 35 days after breeding. During the postpartum period, PAG decrease steadily and this assay can be used in cows 60 days and older after calving.
THE SCOPE OF USE
The test is mainly suitable for dairy and beef cattle early pregnancy diagnosis, as well as to guide timely artificial insemination.
STORAGE AND STABILITY
Store as packaged in the sealed pouch either at room temperature or refrigerated (2-30 °C). The test is stable through the expiration date printed on the sealed pouch. The test cassette must remain in the sealed pouch until use. DO NOT FREEZE. Do not use beyond the expiration date.
PRECAUTIONS
Do not use after the expiration date.
Handle all specimens as if they contain infectious agents. Observe established precautions against microbiological hazards throughout testing and follow the standard procedures for proper disposal of specimens.
Wear disposable gloves and eye protection when specimens are being tested.
Humidity and temperature can adversely affect results.
Do not remove test cassette from its pouch until immediately before use.
Do not reuse the test kit.
Do not mix components from different lot and different products.
DIRECTIONS FOR USE
Allow the test cassette, specimen, buffer to equilibrate to room temperature (15-30 °C) prior to testing.
1. Remove the test cassette from the foil pouch and use it as soon as possible. Best results will be obtained if the assay is performed within one hour.
2. Place the test cassette on a clean and level surface. Hold the dropper vertically and transfer 1 drop of serum or plasma (approximately 25 μL) to the specimen well (S) of the test cassette, then quickly add 2 drops of buffer (approximately 80 μL) and start the timer. See illustration below.
3. Read the results at 10 minutes. Do not interpret results after 20 minutes.


Bovine Pregnancy (PAG) Rapid Test Cassette has been evaluated with specimens obtained from one dairy farm. ELISA is used as the reference method for the Bovine Pregnancy (PAG) Rapid Test Cassette. Specimens were considered positive if ELISA indicated a positive result.
